Member states will have an opportunity to identify challenges, recommend strategies and review progress and map the future course of the global Aids response.
The gathering will also be a platform for countries to recommit to a sustainable response to HIV/Aids, broader health issues and the development agenda.
Motlanthe will be accompanied by Health Minister Dr Aaron Motsoaledi other senior government officials.
